Respiratory illness are complex human diseases, most therapeutic and pathological understandings of these diseases are learned from animal models. Primates have long provided as research models for studying complexities of respiratory diseases due to sharing similar genetic and physiological patterns with human, as well as their large size of respiratory system, which permit histological analysis and bronchoscopy examination.
Allergic asthma is a chronic inflammatory disease caused by allergens and usually results in airway dysfunction such as breathlessness, wheezing and coughing. Mouse is mostly used for allergic asthma model due to low cost and easy sensitization. However, compared to human, mouse with allergic asthma rarely displays inflammatory mucosa and infiltrating of eosinophilic cell in upper cortex of airway. Moreover, primate model has preponderance of similar physiological lung structure to human. Creative Animodel provides allergic asthma model in rhesus and cynomolgus monkey by human allergens such as dust mite, pollen allergen and other parasites. Monkey allergic asthma model is initiated by sensitivity test via cell counts of bronchoalveolar lavage fluid and monkeys has high allergic risk are selected. Allergen-sensitive animal will receive allergen challenge via nebulized, intratracheal or intranasal administration routes. For parasite extract or antigen, subcutaneously injection of alum adsorbed allergens also are applicable.
Creative Animodel offers complete set of following analyzing serves for allergic asthma model. These services include but not limit to pulmonary function tests, ELISA for allergen and cytokine detection, pathological analysis. Pulmonary function is measured by inserting endotracheal tube and esophageal balloon linked to computer. Chronic Obstructive Pulmonary Disease (COPD)
COPD is considered as one of leading cause of death diseases and has drawn many researchers’ attentions. Compared to small animals, Creative Animodel established primate COPD model can overcome limitations such as different pulmonary anatomy (monopodial airway in mice and dichotomous pattern in human), lack of mucus hypersecretion and small airway fibrosis, and immune responses. Cigarette smoke-exposed (CS) primate model is developed in Creative Animodel for COPD lung inflammation. Primate, especially in cynomolgus and rhesus monkey are recommended for this model. Primates are exposed to cigarette smoke in either whole body chamber or mainstream (head only) apparatus, and followed by well-proved protocol. Smoke dose in CS system can be defined by total suspended particles and total particle matter, and depended on type of cigarette, additional references can be included. Creative Animodel also provide lipopolysaccharide (LPS)-induced COPD inflammation as separate or combinative part of CS model. Animal receive LPS via inhalation during CS exposure or inject intratracheally. Other COPD features such as airspace enlargement and emphysema also can be induced by chronic CS treatment (about 6 months).
