Humanized-Xenograft Models

One crucial shortcoming of PDX models is the insufficient immune function, which makes it impossible to perform the efficacy tests of any immunotherapies or anti-cancer agents that target the immune system components. As a consequence, researchers are beginning to develop humanized-xenograft models by co-engrafting the patients` tumor fragments and peripheral blood or bone marrow cells into a NOD/SCID mouse or NSG mouse, which allows for the reconstitution of the murine immune system enabling the researches on the interactions between human stroma and tumor environments in cancer progression and metastasis.

To actively meet the challenges of scientific research, Creative Animodel have established two well-tested humanized-xenograft models including CD34+ humanized mice and PBMC humanized mice so as to provide a system for testing the inter-reaction between treatments and the human immune system, meanwhile for imitating models of diverse infectious diseases from allergies, inflammation to graft versus host disease.

Humanized-Xenograft Models

• CD34+ Humanized Mice

Creative Animodel provides humanized CD34+ mouse models which engrafted with cord blood-derived hematopoietic stem cells into highly immune-deficient NSG mice, which enables long-term studies involved in immunology-related researches such as graft versus host disease. Generally, the percentage of human CD45+ cells could reach 25% after a 12-week period of engraftment. Models of this type efficiently develop a functional human immune system which allowing T-cell maturation as well as its associated immune responses.   

• PBMC Humanized Mice

Meanwhile, Creative Animodel provides alternative humanized mice species generated by engraftment of human peripheral blood mononuclear cells (PBMC) into highly immune-deficient NSG mice. Hu-PBMC models present a shorter engraftment period of 3-4weeks that enabling the summit of lymphocytes proliferation compared with humanized CD34+ models. This model type is thereof fit for short-term studies requiring a strong effector and memory T cell and NK cell function such as evaluating therapeutics for infectious diseases.

• Immuno-oncology

The strategy by pharmacologically stimulating patients’ immune responses to destroy tumor cells (immuno-oncology) can be achieved by drug intervention on the patient’s immune system, which possess the advantages to avoid off-target toxicities as well as undesired side effects comparing to the traditional chemotherapeutics.
