In Vitro Hematotoxicity Testing


Stem cells in hematopoietic system illustrate the potential to predict partial or life-threatening damage in response to drugs, environmental compounds and other agents. In vitro assay of detecting stem cell toxicity in a biological system allows potential prediction of life-threatening damage in pre-clinical studies. Creative Animodel has carried out the in vitro hematotoxicity testing platform to study the toxicity of drugs or compound to the blood system in pre-clinical animal testing by using the colony-forming cell (CFC) assay.

In Vitro Hematotoxicity Testing

Assays for in vitro hematotoxicity testing:
Three different assays for the detection and measurement of hematopoietic stem cells (HSC), progenitor cells (HPC) and precursor cells have been conducted in Creative Animdoel in the following:
● Integration of 96-well and 384-well plate assay is applied for toxicity studies of all HSCs and HPCs.
● 96-well plate assay is utilized where both cell proliferation and differentiation response to compound administration have to be measured.
● 4-well plate assay is generated for methylcellulose colony-forming cell to better understand the effect of compounds on the differentiation and maturation process of hematopoietic development.

Available species for hematotoxicity testing:
● Non-human primate (such as macaca fascicularis, cynomologus (Cyno) and Macaca mulatta (Rhesus))
● Horse
● Swine
● Sheep
● Canine
● Murine (including multiple strains of rat and mouse)

Cells and tissues used for hemototoxicity testing:
● Bone marrow of almost all species
● Peripheral blood for human and large animals
● Umbilical cord blood which is only for human
● Purified stem cell populations such as human CD34+ and CD133+ cells
● Hematopoietic organs from animals such as spleen, fetal liver and yolk sac.
