What Is Isotopic Labeling?

Isotopic labeling is an analytic technique that is used to track the movement of isotopes through reaction networks. For isotopic labeling technology, a specific atom of the reagent moieties is replaced with labeled isotopic variant, then, the position and relative abundance of labeled isotopic atoms are determined by experiment. Subsequent analysis of the measured quantities elucidates the characteristics of the reaction network. The experiment flow of 13C-labeling technique is shown in figure 1, it provides a means to simply and directly assess the bio-behavior of unaltered carbon nanomaterials (including fullerene), which would largely benefit evaluation of the biosafety and environmental fate studies of carbon nanomaterials.

An injection of a radioisotopically labeled drug can be used to determine blood, plasma, or urine levels and their temporal profile by liquid scintillation or gamma counting and measurement of activities in serially derived samples. Liquid scintillation counters can be used to measure total sample radioactivity. In addition, radioactivity of individual components can be determined after chromatographic separation by using an online radiometric chromatographic detector or by collecting chromatographic fractions and using a liquid scintillation detector. Scientists can trace the detailed process of chemical reactions by isotopic labeling, which has been widely used in the clinical research of medical field.

Types of Isotopic Labeling

▪ Uniform labeling with the stable isotopes 13C, 15N, and 2H
Target-based NMR screening relies on the detection of perturbations of target resonances on ligand binding. The most well-known technique is the SAR by NMR method. Ligands are identified by monitoring alterations of target signals in a 2D 1H–15N correlation spectrum. This method requires large quantities of isotopically (13N)-labeled protein (50–200 mg, with protein solubility between 0.1 and 1 mM). This method has the benefit (provided that sequence-specific resonance assignments have been obtained) of high reliability, being able to identify the binding site on the target, readily distinguishing specific from nonspecific interactions, and assessing whether any significant conformational changes occur upon binding. The incorporation of 13C into the target protein may also be used for the detection of binding of small molecules to targets.
