Why Ultrafiltration Technique?
Plasma protein binding (PPB) plays an important role in the pharmacokinetics (PK) and pharmacodynamics (PD) of a drug. Free drug levels are closely related to the pharmacological effects of a compound which is as the principal determinant of tissue distribution, cell entry, receptor interactions, and effective elimination. At the early stages of drug development, PPB experiments advance understanding of ADME properties to aid in candidate selection. Ultrafiltration (UF) technique is an easy-operating and time-saving approach with the centrifugal force to separate free drug from that bound to plasma proteins through a size exclusion membrane. UF that relies on convection (or pressure)-driven transport of drug molecules along with solvent through a membrane is faster and does not lead to appreciable change of the free drug concentration in the donor compartment.

Our PPB Assay by Ultrafiltration
Creative Animodel offers PPB assays by ultrafiltration for a wide range of species and strains. Our experts have been attempting to reduce non-specific binding (NSB) by the use of alternative membrane materials or chemical pretreatment through the UF device. A novel and practical sample pretreatment method based on hollow fiber centrifugal ultrafiltration is also developed to determine plasma protein binding by using HPLC at Creative Animodel. Hollow fiber centrifugal ultrafiltration (HFCF-UF), a simple device composed of a slim glass tube and a U-shaped hollow fiber, has been introduced for the separation of unbound drug fraction from plasma sample. Our PPB assays are accomplished by following three steps: incubation, ultrafiltration and HPLC-UV analysis.
