Cell Viability and Proliferation

The determination of cell viability and proliferation is important in fields involving cell-relevant studies. The cell viability is to assess how healthy the cells are, which is necessarily evaluated prior to any cell-based study. And the quantification of cellular proliferation is used to determine optimal culture conditions, as well as to investigate the cytotoxic effect mediated by the treatment of choice. We provide both sulforhodamine B (SRB) assay in an efficient manner, and MTT assay in a high-throughput and sensitive manner for the evaluation of cell viability and proliferation.

Our Cell Viability and Proliferation Assays

SRB assay
The SRB assay has been widely performed to detect cytotoxicity in cells. Compared to MTT assay, this method does not measure metabolic activity, therefore it is highly cost-effective and simplified. SRB assay relies on the property of SRB, which binds stoichiometrically to proteins under mild acidic conditions and then can be extracted using basic conditions. Apart from acting as a proxy for cell mass, SRB assay can also be used to evaluate gene expression modulation, as well as the effects of miRNA replacement on cell proliferation.
MTT assay
MTT assay is a colorimetric method for measuring the activity of enzymes in living cells that give a purple color by reducing MTT to formazan dyes. For example, the yellow tetrazolium MTT is reduced by the action of dehydrogenase enzymes, hence to cause reduced equivalents such as NADH and NADPH. The resulting formazan can be solubilized and quantified by MS platforms. When metabolic events generate apoptosis or necrosis, the cell viability rate decreases. Therefore, MTT assay is commonly performed to determine the potential cytotoxicity mediated by test medicinal agents and other substances.
