What Is Ischemia?
Ischemia occurs when there isn’t enough supply of blood to tissues or organs, which is usually due to the problems with blood vessels, resulting in poor oxygen supply for cells, further causing unconsciousness, coordination disorder, and even blindness, stroke, cardiorespiratory arrest, etc. Ischemic stroke is the third most common disease-related killer in the United States. Besides, the lesion produced by the ischemia will result in both acute and chronic allodynia (in some cases) after mechanical and cold stimulation. It’s necessary for us to use animal models to mimic human ischemic injury for the ischemia in vivo studies.
Depending on the location of the occurrence, ischemia can be divided into global ischemia and focal ischemia. Global ischemia occurs when all blood supply to the brain is interrupted or markedly reduced. This can be induced by intrauterine clamping of the carotid arteries or the umbilical cord. Focal ischemia affects one or more specific regions as defined by the vascular anatomy and is characterized by induction of infarctions within these regions.
Pharmacology Evaluation in Ischemia Models
Creative Animodel conducts both global ischemia and focal ischemia models to study ischemia and provides a wide range of drug efficacy evaluation services. Metabolic and physiological parameters are measured before, during, and after ischemia. Parameters include the plasma glucose concentration, arterial blood pressure, and pH, heart rate, etc. Arterial blood pressure and heart rate are monitored via an intra-arterial catheter in the right femoral artery, which is connected to a multichannel recording system.
• Global Ischemia Models
The global ischemia models can be differentiated into total-body ischemia, global cerebral ischemia, and forebrain ischemia.

• Focal Ischemia Models
The most important focal ischemia models include proximal main carotid artery occlusion, intravascular thread, distal main carotid artery occlusion with bilateral common carotid artery (CCA) occlusion, tandem occlusion and photothrombosis. The transient middle cerebral artery occlusion (MCAO) animal model is also one of the widely used focal ischemia models.
