Lipopolysaccharide (LPS), a structural part of the outer membrane of Gram-negative bacteria, is one of the most effective stimulators of the immune system and has been widely applied as an experimental model for bacterial infection.
Acute phase response (APR) is a group of physiologic processes occurring soon after the onset of infection, trauma, inflammatory processes, and some malignant conditions. The most prominent change is a dramatic increase of acute phase proteins in the serum, especially C-reactive protein. Also seen are fever, increased vascular permeability, and a variety of metabolic and pathologic changes.
Models of acute response to LPS challenge can be used to investigate the effect of targeting mechanisms related to neutrophil recruitment and mediator release, such as acute bacterial infection, acute inflammation, adult respiratory distress syndrome (ARDS), cystic fibrosis (CF) and chronic obstructive pulmonary disease (COPD).
LPS Acute Response Models Provided by Creative Animodel
Different types of animals can be used to establish acute response model by LPS induced, e.g. mice, rats, rabbits, and pigs. In general, animals need to be injected intraperitoneally (i.p.) with a dose of LPS to develop acute phase response (mice and rats can be exposed to an airway of LPS via the intranasal route to induce acute response). But the selected E. coli serotype and LPS dose, the duration and route of administration (ROA) are important variables to these models established.
